目的应用单激光流式细胞仪检测法,建立快速、自动分析骨髓红细胞微核率的检测方法。方法以秋水仙碱诱导雄性Wistar大鼠微核形成,采用吖叮橙(AO)荧光染色,分别用荧光显微镜及单激光流式细胞仪检测大鼠骨髓嗜多染红细胞微核率和正染红细胞微核率。结果随着秋水仙碱浓度的增加,大鼠骨髓嗜多染红细胞微核率和正染红细胞微核率也随之增高,有良好的量效相关性;人工显微镜计数结果与流式细胞仪检测结果比较分析表明,两种方法的实验结果具有良好的相关性(r=0.958,P<0.01)。结论单激光流式细胞仪可替代人工方法检测骨髓红细胞微核率,方法快速、稳定。
Objective To establish a rapid and automatic method for the detection of micronuclei in erythrocytes of bone marrow by single laser flow cytometry. Methods Micronucleus formation in male Wistar rats was induced by colchicine. Fluorescence microscopy and single laser flow cytometry were used to detect micronucleus rate and positive cell number of erythrocytes in rat bone marrow using acridine orange (AO) staining. Nuclear rate. Results With the increase of colchicine concentration, micronucleus rate of erythrocytes and micronuclei of positive erythrocytes in rat bone marrow also increased, and there was a good dose-effect correlation. The counting results of artificial microscope and flow cytometry The comparative analysis shows that the experimental results of the two methods have a good correlation (r = 0.958, P <0.01). Conclusion Single laser flow cytometry can replace the manual method to detect micronuclei of bone marrow erythrocytes, the method is rapid and stable.
