[目的]优化福鼎大白茶愈伤组织的诱导条件。[方法]以茶叶品种福鼎大白茶的幼嫩茎段为外植体,以MS为基本培养基,添加不同浓度的2,4-D、KT、NAA和蔗糖,研究不同激素组合、蔗糖浓度对其愈伤组织诱导的影响。在继代培养过程中,研究了不同抗褐变剂对愈伤组织生长情况的影响及抗褐变的效果。[结果]不同激素对福鼎大白茶愈伤组织的诱导影响依次为2,4-D>KT>NAA。适宜福鼎大白茶愈伤组织生长的培养基为MS+2,4-D1.0 mg/L+NAA1.0 mg/L+KT0.5 mg/L+蔗糖30 g/L,pH值为5.8。在培养基中添加0.5%的活性炭和0.1%的聚乙烯吡咯烷酮可有效减轻愈伤组织的褐变。[结论]不同的激素组合、蔗糖浓度及抗褐变剂对福鼎大白茶愈伤组织的诱导均有较大影响。
[Objective] The research aimed to optimize the induction conditions of Fuding Dabaicao callus. [Method] The young shoots of Fuding Dacha were used as explants with different concentrations of 2,4-D, KT, NAA and sucrose as basal medium. The effects of different hormone combinations, sucrose concentration On callus induction. In the process of subculture, the effects of different anti-browning agent on the growth of callus and the anti-browning effect were studied. [Result] The effects of different hormones on callus induction of Fuding Da Bai Cha were 2,4-D> KT> NAA. The medium suitable for the growth of Callus of Fuding dabai tea was MS + 2,4-D1.0 mg / L + NAA1.0 mg / L + KT0.5 mg / L + sucrose 30 g / L and pH 5.8. Adding 0.5% activated carbon and 0.1% polyvinylpyrrolidone to the medium can effectively reduce browning of callus. [Conclusion] Different hormonal combinations, sucrose concentration and anti-browning agents had a significant effect on the induction of callus of Fuding Da Bai Cha.
