为建立基于样品DNA快速制备、PCR-LFB的羊源性成分检测技术,为动物源性成分的检测提供新思路.进行动物源性成分DNA快速提取,针对绵羊线粒体基因组序列设计基因特异性引物及探针进行PCR扩增,制备LFB对羊源性成分核酸扩增物进行检测.结果表明:样品DNA快速制备技术操作简单、耗时少;PCR-LFB对羊基因组DNA检测灵敏度为常规琼脂糖凝胶电泳的10倍,特异性为100%,对9种商业化羊肉样品均可检出.建立的羊源性成分PCR-LFB检测技术,具有简单、快速、准确的特点.“,”A detection method for sheep-derived ingredients based on rapid DNA extraction and PCR-lateral flow biosensor was established, which will provide new ideas for detecting animal derived ingredients. DNA was extracted from animal materials and gene specific primers and probes were designed to amplify ovine mitochondrial genome sequence; lateral flow biosensor( LFB) was used to analyze nucleic acid of the amplified sheep-derived ingredients rapidly. In this study, the DNA extraction operation was simple and less time consuming. The sensitivity of PCR-LFB to the detection of sheep-derived ingredients was 10 times higher than that of conventional agarose gel electrophoresis, and the specificity was 100%. The method was applied to detect nine commercial mutton products. This study has established the PCR-lateral flow biosensor detection technology for sheep-derived ingredients, which has the characteristics of simplicity and accuracy, and can be used for carrying out rapid detection of sheep-derived ingredients at the primary-level laboratory.
