目的 研究蛇床子素对成骨细胞功能的影响。方法 原代培养新生大鼠颅盖骨成骨细胞 ,3H 胸腺嘧啶掺入法测定成骨细胞的增殖 ;Griess试剂法测定培养上清中NO的含量 ;小鼠胸腺细胞增殖法测定IL 1的含量 ;B9细胞增殖法测定IL 6的含量 ;RT PCR法检测IL 6的mRNA的表达。结果 蛇床子素抑制成骨细胞自发地或在炎性细胞因子及LPS刺激下产生NO、IL 1及IL 6和IL 6mRNA表达。结论 蛇床子素通过减少NO、IL 1和IL 6的分泌而调节成骨细胞的功能
Objective To study the effect of osthole on osteoblast function. Methods Primary rat calvarial osteoblasts were cultured, osteoblast proliferation was measured by 3H thymidine incorporation method, NO content in culture supernatant was measured by Griess reagent method, and IL 1 content was determined by mouse thymocyte proliferation assay. B9 cell proliferation method was used to determine the content of IL 6; RT PCR method was used to detect the expression of IL 6 mRNA. RESULTS Osthole inhibited the production of NO, IL 1, IL 6 and IL 6 mRNA by osteoblasts spontaneously or stimulated by inflammatory cytokines and LPS. Conclusion Osthole regulates the function of osteoblasts by reducing NO, IL 1 and IL 6 secretion
