目的观察青秦液对高尿酸血症模型大鼠关节免疫性病理损伤的修复作用。方法将实验动物分为正常组、模型组、阳性药组及青秦液大、中、小剂量组,采用灌服腺嘌呤及乙胺丁醇进行高尿酸血症造模。阳性药组按0.09mg/kg体重灌服秋水仙碱;青秦液大剂量组给予5g/(kg·d)的青秦液灌胃,中剂量组给予2.5g/(kg·d)青秦液灌胃;小剂量组给予1.25g/(kg·d)灌胃。1周后,在双侧踝关节背侧将尿酸钠溶液注射到关节腔中。72h后处死动物,抽取关节腔滑液,应用ELISA法检测大鼠关节腔滑液中的炎性细胞因子白细胞介素-8(IL-8)、α-肿瘤坏死因子(TNF-α)的水平,并取踝关节固定于10%福尔马林中,脱钙后常规石蜡包埋,HE染色,光镜观察其用药前后关节周围炎性细胞浸润情况。结果阳性药组、青秦液3个剂量组大鼠关节腔滑液中炎性细胞因子IL-8、TNF-α与模型组比较差异均无统计学意义(P>0.05),但从数据上可以看出,青秦液小剂量组大鼠关节腔滑液中IL-8含量低于模型组、阳性药组及青秦液大、中剂量组,而青秦液3个剂量组大鼠关节腔滑液中TNF-α含量低于模型组及阳性药组,其中尤以青秦液中剂量组为低。光镜观察各用药组关节滑膜增生、周围软组织炎症及关节囊渗出均较模型组减轻。结论青秦液对高尿酸血症大鼠急性反应性关节炎病理损伤具有一定的修复作用。
Objective To observe the effect of Qingqin liquid on the repair of joint immunopathological injury in hyperuricemia model rats. Methods The experimental animals were divided into normal group, model group, positive drug group and large, medium and low dose group of Qingqin fluid. Auroral and ethambutol were used to establish hyperuricemia model. The positive drug group was fed with 0.09 mg/kg body weight of colchicine; Qingqin high-dose group was given 5g/(kg·d) of Qingqin liquid, and the middle dose group was given 2.5g/(kg·d) of Qingqin. Liquid gavage; small dose group given 1.25g/(kg · d) gavage. One week later, sodium urate solution was injected into the joint cavity on the dorsal side of bilateral ankle joints. After 72 hours, the animals were sacrificed and the synovial fluid was extracted from the joint cavity. The levels of interleukin-8 (IL-8) and α-tumor necrosis factor (TNF-α) in synovial fluid of rats were measured by ELISA. The sacroiliac joints were fixed in 10% formalin. After decalcification, they were embedded in paraffin and stained with HE. The infiltration of inflammatory cells around the joints was observed by light microscopy. Results There were no significant differences in inflammatory cytokines IL-8 and TNF-α between synovial fluid from the positive drug group and Qingqin liquid group and the model group (P>0.05), but from the data It can be seen that the content of IL-8 in synovial fluid of rats in the low dose group of Qingqin liquid is lower than that in the model group, the positive drug group, and the large and medium dose group of Qingqin fluid, while the Qingqin fluid in the three dose groups of rat joints The content of TNF-α in the synovial fluid was lower than that in the model group and the positive drug group, especially in the middle dose group of Qingqin liquid. Observed by light microscope, synovial hyperplasia, peripheral soft tissue inflammation and joint sac leakage in each drug group were reduced compared with the model group. Conclusion Qingqin liquid can repair the pathological injury of acute reactive arthritis in hyperuricemia rats.
