目的:研究翘芩清肺剂对哮喘模型大鼠的平喘作用及其与血小板活化因子(PAF)、白细胞总数(WBC)、嗜酸性粒细胞(EOS)及内皮素(ET)含量的变化关系,以探讨其作用机制。方法:将大鼠分为正常对照组、哮喘模型组、翘芩清肺剂高、中、低剂量组、地塞米松组和氨茶碱组,以卵白蛋白(OVA)建立哮喘大鼠模型。除正常组外,其余各组均在第1,8天以10%OVA 1 mL ip致敏,第15天起雾化吸入1%OVA激发哮喘,每天1次,每次20 min,连续2周,建立哮喘模型。从大鼠致敏第2天开始,各组大鼠以对应药物ig给药(其中地塞米松组从哮喘激发当日开始)。正常组、模型组给生理盐水5 mL.kg-1.d-1,地塞米松组0.27 mg.kg-1.d-1,氨茶碱组50 mg.kg-1.d-1,翘芩清肺剂高、中、低剂量组26,13,6.5 g.kg-1。造模时间为4周,取材后处死大鼠,观察药物对病鼠的哮喘程度、哮喘潜伏期的影响;测定血清PAF,WBC,EOS及肺泡灌洗液(BALF)中ET含量的变化。结果:翘芩清肺剂能有效减轻哮喘模型的哮喘程度(P<0.05)及明显延长病鼠的哮喘潜伏期(P<0.01),降低病鼠的PAF,WBC,EOS含量及ET水平,与模型组比较有显著性差异(P<0.05或P<0.01)。结论:翘芩清肺剂可有效改善哮喘状态,减轻哮喘发作,其作用机制可能与降低PAF,WBC,EOS及ET水平密切相关。
Objective: To investigate the anti-asthmatic effect of “Qiyan qingfei decoction” on asthma in rats and its relationship with the changes of platelet activating factor (PAF), total white blood cell (WBC), eosinophil (EOS) and endothelin (ET) , To explore its mechanism of action. Methods: The rats were divided into normal control group, asthma model group, high, medium and low dose of QQL, dexamethasone group and aminophylline group. Asthma rat model was established by ovalbumin (OVA). Except the normal group, all other groups were sensitized with 10% OVA 1 mL ip on days 1 and 8, and inhaled 1% OVA aerosolized on the 15th day to stimulate asthma once a day for 20 min each for 2 weeks , Establish asthma model. Starting from day 2 of rat sensitization, rats in each group were dosed with the corresponding drugs (dexamethasone group started on the day of asthma challenge). Normal group, the model group to saline 5 mL.kg-1.d-1, dexamethasone group 0.27 mg.kg-1.d-1, aminophylline group 50 mg.kg-1.d-1, Alice Qin Qingfei agent high, medium and low dose group 26,13,6.5 g.kg-1. The model was established for 4 weeks. The rats were sacrificed and the effects of the drugs on the degree of asthma and the latent period of asthma were observed. The contents of ET in serum PAF, WBC, EOS and BALF were measured. Results: The results showed that “Qqq” could effectively reduce the degree of asthma (P <0.05) and prolong the latent period of asthma in asthmatic mice (P <0.01), decrease the content of PAF, WBC, EOS and the level of ET, There was significant difference between the two groups (P <0.05 or P <0.01). Conclusion: QQQY can effectively improve the state of asthma and reduce the incidence of asthma. Its mechanism may be related to the reduction of PAF, WBC, EOS and ET levels.
