枯草芽孢杆菌CG24是一株烟草野火病菌拮抗菌,本文采用单因素实验和响应面法对该菌株发酵条件进行了优化。结果表明CG24菌株在培养温度32℃初始pH 7.5装液量50 mL/150 mL和接种量4%的NA培养液中振荡培养28 h,其代谢产物的抑菌活性作用最为显著。通过单因素实验确定了影响菌株CG24生长的培养基主要因素为葡萄糖、酵母膏和氯化钙,对3个因素进行响应面优化分析,确定最优发酵培养基为:葡萄糖40 g/L酵母膏7.55 g/L氯化钙0.72 g/L,抑菌圈直径比优化前提高了25.72%,为该菌株以后的扩大培养和微生态制剂的开发应用奠定了基础。
Bacillus subtilis CG24 is a antagonistic bacterium of wild tobacco. In this paper, the fermentation conditions were optimized by single factor experiment and response surface methodology. The results showed that the antibacterial activity of CG24 strain was the most significant when the CG24 strain was cultured in NA culture medium containing 50 mL / 150 mL of initial pH 7.5 and 4% inoculum at 32 ℃ for 28 h. Through the single factor experiment, the main factors affecting the growth of strain CG24 were glucose, yeast extract and calcium chloride, and the response surface optimization analysis of three factors was carried out. The optimal fermentation medium was glucose 40 g / L yeast extract The diameter of bacteriostasis ring increased by 25.72% compared with the pre-optimization 7.55 g / L CaCl2 0.72 g / L, which laid the foundation for further expansion of this strain and the development and application of probiotics.
