目的 :研究不同期哮喘患者外周淋巴细胞 β肾上腺素能受体数量及功能的变化。 方法 :用放射配基结合分析及放射免疫分析法测定外周淋巴细胞 β肾上腺素能受体和血浆cAMP、cGMP水平 ,同时测定通气功能。结果 :缓解期哮喘患者外周淋巴细胞 β肾上腺素能受体最大结合量 (Bmax)较正常人明显增高 ,平衡解离常数 (Kd)亦增高 ,血浆cAMP水平无显著差异 ;发作期哮喘患者 β肾上腺素能受体Bmax较正常人显著降低 ,Kd值无明显差异 ,而血浆cAMP水平则显著降低。结论 :哮喘时 β肾上腺素能受体的改变与哮喘患者所处状态有关 ,是其发病过程中伴随出现的病理变化。
Objective: To study the changes of the number and function of β-adrenergic receptors in peripheral lymphocytes in patients with asthma at different stages. Methods: Radiolipoprotein binding assay and radioimmunoassay were used to determine the level of cAMP and cGMP in the peripheral blood lymphocytes and β-adrenoceptors. Ventilation function was also measured. Results: The maximal binding capacity of β-adrenergic receptor in peripheral lymphocytes in asthma patients was significantly higher than that in normal subjects (P <0.05), and the equilibrium dissociation constant (Kd) was also increased. There was no significant difference in plasma cAMP levels between the two groups The Bmax of erythropoietin receptor was significantly lower than that of normal subjects, but there was no significant difference between Kd values and plasma cAMP level. Conclusion: The changes of β-adrenergic receptor in asthma are related to the state of asthma patients and the pathological changes accompanying the pathogenesis of asthma.
