目的研究神经生长因子(NGF)对实验性视网膜脱离(RD)后视网膜细胞凋亡相关基因c-fos、bcl-2、Fas-L与caspase-3表达的影响。方法20只大鼠右眼和左眼分别设为实验组(NGF组)和实验对照组(PBS组),另设立正常对照2只。通过在视网膜下注射透明质酸钠的方法建立视网膜脱离(RD)动物模型后,右眼在玻璃体腔内注射NGF 5μg/次(1 g/L),每4 d注射1次,作为NGF组;左眼注射PBS作为实验对照组。两组在建立模型后1.5、3、6、12 h和1、2、4、8、16、32 d分别取眼球,采用免疫组化学法对视网膜细胞的bc l-2、Fas-L、caspase-3和c-fos进行标记。结果Fas-L、bc l-2、caspase-3和c-fos在实验各组均有表达,并随着时间的变化而变化。NGF组的各基因蛋白表达与PBS组相比差异有统计学意义(P<0.05)。NGF组的c-fos蛋白表达与PBS组相比,表达高峰时间延迟,且表达量下降(P<0.05)。结论实验性RD后给予外源性NGF能促进bc l-2基因的表达,抑制c-fos基因和Fas-L的表达,影响caspase-3的激活和表达,从而抑制RD后视网膜细胞凋亡的发生。
Objective To investigate the effects of nerve growth factor (NGF) on the expression of apoptosis-related genes c-fos, bcl-2, Fas-L and caspase-3 in retina after retinal detachment (RD) Methods Right eyes and left eyes of 20 rats were randomly divided into experimental group (NGF group) and experimental control group (PBS group). Two normal control subjects were set up. The retinal detachment (RD) animal model was established by subretinal injection of sodium hyaluronate. The right eye was intravitreally injected with 5 μg / L NGF (1 g / L) and injected once every 4 days as the NGF group. Left eye PBS was injected as experimental control group. The two groups were enucleated at 1.5, 3, 6, 12 h and 1, 2, 4, 8, 16 and 32 d after establishment of the model respectively. Immunohistochemistry was used to detect the expression of bcl-2, Fas-L, caspase -3 and c-fos. Results Fas-L, bcl-2, caspase-3 and c-fos were all expressed in the experimental groups and changed with time. Compared with PBS group, the expression of each gene in NGF group was statistically significant (P <0.05). The expression of c-fos protein in NGF group was delayed compared with PBS group, and the expression of c-fos protein was decreased (P <0.05). Conclusions Exogenous NGF can promote the expression of bcl-2 gene, inhibit the expression of c-fos gene and Fas-L, and affect the activation and expression of caspase-3, thereby inhibiting the apoptosis of RD retinal cells occur.
