目的观察葛根素对动脉粥样硬化兔髂动脉分泌和表达C反应蛋白(CRP)的影响。方法20只家兔分为正常对照组(6只,正常饮食)、基础对照组(8只,球囊损伤和高脂饮食)和葛根素干预组(8只,球囊损伤、高脂饮食和葛根素干预),球囊损伤后4周处死;取一侧病变髂动脉做病理切片,应用免疫组化法测定CRP的表达;取另一侧病变髂动脉抽提总RNA,应用半定量逆转录多聚酶链式反应(RT-PCR)测定CRP mRNA的表达。结果葛根素干预组髂动脉粥样斑块CRP mRNA及其蛋白质表达均明显低于基础对照组(P<0.01)。结论葛根素可以通过调节兔动脉粥样斑块分泌CRP途径发挥稳定动脉粥样硬化斑块的作用。
Objective To observe the effect of puerarin on the secretion and expression of C-reactive protein (CRP) in the iliac artery of atherosclerotic rabbits. Methods Twenty rabbits were divided into normal control group (n = 6), normal control group (n = 8), balloon injury and high fat diet and puerarin intervention group (n = 8) Puerarin intervention), 4 weeks after balloon injury were sacrificed; take iliac artery from one side to make pathological sections, and the expression of CRP was detected by immunohistochemical method; total RNA was extracted from the iliac artery on the other side and semi-quantitative reverse transcription CRP mRNA expression was determined by polymerase chain reaction (RT-PCR). Results The expression of CRP mRNA and protein in iliac artery plaque in the puerarin intervention group was significantly lower than that in the control group (P <0.01). Conclusion Puerarin can stabilize the atherosclerotic plaque by regulating CRP secretion in atherosclerotic plaque in rabbits.
