目的 探讨间接同种异型识别在体外培养的同种异体表皮细胞移植排斥反应中的作用。方法 体外分离培养人表皮细胞 (Epidermalcells ,EC)、异体人外周血淋巴细胞 (Peripheralbloolymphocytes ,PBL)和单个核细胞 (Peripheralbloodmononuclearcells ,PBM ,含单核细胞 )。将毒性T淋巴细胞相关抗原 4(CTLA4Ig)腺病毒载体转染EC。转染与未转染的EC中均加入异体PBL或PBM共同培养 ,应用氚标记胸腺嘧啶脱氧核苷 ( 3H TdR)掺入法 ,测定各培养系中PBL、PBM增殖情况。结果 CTLA4Ig腺病毒载体能成功转染EC并表达相应蛋白。未转染的EC刺激异体PBL轻度增殖 (P <0 .0 5 ) ,以及刺激含单核细胞的PBM明显增殖 (P <0 .0 1) ;经CTLA4Ig腺病毒载体转染的EC刺激PBL、PBM增殖则明显减弱 (P <0 0 5 )。结论 间接同种异型识别在EC排斥中发挥了重要作用 ,CTLA4Ig能明显减轻该作用。
Objective To investigate the role of indirect allotyping in allogeneic epidermal cell transplantation rejection in vitro. Methods Epidermal cells (ECs), Peripheral blood mononuclear cells (PBLs) and mononuclear cells (PBMCs) were isolated and cultured in vitro. Toxic T lymphocyte associated antigen 4 (CTLA4Ig) adenovirus vector was transfected into ECs. The transfected and non-transfected ECs were co-cultured with allogeneic PBLs or PBMs. The proliferation of PBLs and PBMs in each culture line was determined by tritiated 3H-thymidine incorporation. Results CTLA4Ig adenovirus vector can successfully transfected EC and express the corresponding protein. Untransfected EC stimulated mild proliferation of allogeneic PBL (P <0.05) as well as stimulation of mononuclear cell-rich PBM (P <0.01); ECs transfected with CTLA4Ig adenovirus stimulated PBL , PBM proliferation was significantly weakened (P <0 05). Conclusion Indirect allotyping plays an important role in EC rejection. CTLA4Ig can significantly reduce this effect.
