目的探讨脂多糖(lipopolysaccharides,LPS)对大鼠血管平滑肌细胞(vascularsmoothmusclecels,VSMC)诱导型一氧化氮合酶(iNOS)基因表达及一氧化氮(nitricoxide,NO)合成的影响。方法细胞培养、RNA印迹分析和亚硝酸盐含量测定。结果LPS是iNOS的强效诱导物,可在转录水平上诱导VSMCiNOSmRNA表达,促进NO的合成,其作用强度与LPS剂量呈正相关;刺激12h后,VSMCiNOS的表达活性最高,24h开始下降。结论LPS对iNOS表达的诱导作用与其浓度和作用时间有关。
Objective To investigate the effects of lipopolysaccharides (LPS) on iNOS gene expression and nitric oxide (NO) synthesis in rat vascular smooth muscle cells (VSMCs). Methods Cell culture, Northern blot analysis and determination of nitrite content. Results LPS was a potent inducer of iNOS, which induced VSMCiNOS mRNA expression at transcriptional level and promoted the synthesis of NO. The intensity of LPS was positively correlated with the dosage of LPS. After 12 h of stimulation, the expression of VSMCiNOS was highest and decreased at 24h. Conclusion The induction of iNOS expression by LPS is related to its concentration and action time.
