目的探究二十一类样本本身的尿酸层次,评测其对十三类尿酸常规监测体系的基质效应。方法借助同位素混合液相色谱关联质谱方法把各个厂家出品的十三类尿酸监测试剂盒同日立7180型自动生化分析器构成常规监测体系,并将此当作评测方式。借助比较同评测方式测算四十份新鲜冷冻血清同二十一类样本。把比较同评测方式尿酸测算成果实施探究,获得Y预估值双边95%可信区间,评测血清同配备物本身的基质效应。结果全部评测样本内五类基质即新鲜或是冰冻类血清本身的样本于全部常规监测体系内都没有发现基质效应。两类校验品只于一类常规监测体系内发生基质效应。五类EQA型样本内除了运用到准确度证明的新鲜血清类样本,其余冰冻血清类样本于很多常规监测体系内发生基质效应。六类质控品内四类于全部常规监测体系内没有基质效应,两类高质质控品表明负基质效应。结论新鲜血清类样本即正确的监测样本,能够于规范物质配备、质控品配备同EQA内运用。部分校验品于非配备体系内很可能有基质效应而出现校验误差,所以于运用这类校验品以前要通过基质效应评测或是方式证明。
Objective To investigate the level of uric acid in twenty-one samples and to evaluate the matrix effects on routine monitoring system of thirteen uric acid. Methods With the aid of isotope-mixed LC-MS method, the thirteen uric acid monitoring kits produced by various manufacturers and the Hitachi 7180 automatic biochemical analyzer form a regular monitoring system, which is regarded as the evaluation method. With the comparison of the same way to measure the evaluation of 40 fresh frozen serum with twenty one samples. To compare the same way to evaluate the results of uric acid measurement to explore, access to bilateral Y 95% confidence interval, evaluation of serum with the matrix itself matrix effects. RESULTS All five samples in the sample, fresh or frozen, did not show any matrix effect in all routine monitoring systems. Two types of test items occur only in one type of routine monitoring system within the matrix effect. In addition to the use of fresh serum samples with accuracy evidence from the five EQA samples, the remaining frozen serum samples undergo matrix effects in many routine monitoring systems. There are no matrix effects in all six of the six control products in all routine monitoring systems. Two types of high quality controls indicate a negative matrix effect. Conclusion The fresh serum sample, the correct monitoring sample, can be used in the standard material preparation and quality control material preparation together with the EQA. Some of the calibrators may have a matrix error in the non-equipped system, so they should be verified by matrix effects or by way of proof before using such calibrators.
