经FBL3肿瘤抗原预激的骨髓基质细胞对小鼠红白血病FBL3肿瘤细胞具有强烈抑制作用,为探讨骨髓基质细胞直接抑瘤作用的机制,我们从骨髓基质细胞经抗原刺激后分泌的细胞因子和表达的各种免疫相关分子入手进行了探讨.通过ELISA检测发现,经FBL3抗原预激的骨髓基质细胞能分泌较高水平的IL-1β和TNF-α和NO.通过FACS分析还发现经GM-CSF诱导成熟的骨髓基质细胞高表达Fas蛋白,而经FBL3肿瘤抗原刺激后细胞高表达FasL.FasL/Fas途径也是非特异性抗肿瘤作用的一个重要机制.进一步采
The bone marrow stromal cells stimulated by FBL3 tumor antigen have a strong inhibitory effect on mouse erythroleukemia FBL3 tumor cells. To investigate the mechanism of direct inhibition of bone marrow stromal cells, we secrete cytokines and expression from bone marrow stromal cells after antigen stimulation. The various immune-related molecules were explored. ELISA test found that FBL3 antigen prepulsed bone marrow stromal cells can secrete higher levels of IL-1β and TNF-α and NO. FACS analysis also found that by GM-CSF Induction of mature bone marrow stromal cells high expression of Fas protein, and high expression of FasL.FasL/Fas pathway after stimulation by FBL3 tumor antigen is also an important mechanism of non-specific anti-tumor effect.
