丙型肝炎病毒 (HCV)非结构蛋白 5A(NS5A)可与多种细胞内蛋白发生结合 ,涉及调节细胞生长、干扰素耐受、脂质代谢和其他细胞内信号转导通路。本实验通过酵母双杂交技术筛选得到一个与NS5A相结合的未知功能蛋白基因 ,命名为NS5ABP37,根据Gen Bank数据库推定蛋白编码序列的信息 ,应用反转录聚合酶链反应 (PCR)扩增出该基因序列。连接入酵母和真核细胞表达载体表达成功 ,并经回交实验证实NS5A与NS5ABP37的结合作用 ,为进一步研究奠定基础。
Hepatitis C Virus (HCV) Non-structural protein 5A (NS5A) binds to a variety of intracellular proteins and is involved in the regulation of cell growth, interferon tolerance, lipid metabolism, and other intracellular signal transduction pathways. In this study, we got a unknown functional protein gene combined with NS5A by yeast two-hybrid screening and named as NS5ABP37. According to the deduced protein coding sequence information of Gen Bank database, we amplified it by reverse transcription polymerase chain reaction gene sequence. The recombinant plasmid was successfully expressed in yeast and eukaryotic cells, and the binding of NS5A to NS5ABP37 was verified by backcrossing experiment, which laid the foundation for further study.
