目的观察益心口服液预处理对心肌缺血大鼠的影响并初步探讨其作用机制。方法利用Langendorff离体心脏灌注装置,结扎Wistar大鼠冠状动脉左前降支建立心肌缺血模型。将大鼠随机分为假手术组、模型组、地尔硫组、复方丹参滴丸组,以及益心口服液大、中、小剂量组,观察各组结扎前后心脏血流动力学的改变,测定各组心肌超氧化物歧化酶(SOD)的活性和丙二醛(MDA)的含量,检测各组心肌组织中腺苷酸(ATP、ADP、AMP)含量以及能荷(EC)的改变。结果①不同浓度的益心口服液预处理均能改善结扎导致的左室收缩压(LVSP)的降低(P<0.05);大、中剂量益心口服液在个别时间点上明显减缓了结扎导致的左室舒张末期压(LVEDP)的抬高(P<0.05),但对冠脉流量(CF)的改善作用不明显。②益心口服液大、中剂量预处理减轻了缺血心肌SOD活性的降低和MDA含量的升高(P<0.05),其作用与地尔硫组和复方丹参滴丸组无显著性差异(P>0.05)。③益心口服液大、中剂量和复方丹参滴丸均能缓解缺血心肌组织中ATP含量的衰减(P<0.05)。结论益心口服液预处理可对抗离体大鼠心肌缺血损伤,且有剂量依从性,其保护作用可能涉及到抗氧化损伤以及对能量的有效保存。
Objective To observe the effect of Yixin oral liquid preconditioning on rats with myocardial ischemia and to explore its mechanism. Methods A rat model of myocardial ischemia was established by ligation of the left anterior descending coronary artery in Wistar rats using a Langendorff in vitro perfusion device. Rats were randomly divided into sham operation group, model group, diltiazem group, compound Danshen Dripping Pills group, and Yixin oral liquid in large, medium and small dose groups. Changes in cardiac hemodynamics before and after ligation were observed in each group. The activity of superoxide dismutase (SOD) and the content of malondialdehyde (MDA) in myocardium were measured. The contents of adenylate (ATP, ADP, AMP) and the change of energy and load (EC) in myocardium were determined. . Results 1 Pretreatment with different concentrations of Yixin Oral Liquid could improve the reduction of left ventricular systolic pressure (LVSP) induced by ligation (P<0.05); Large and middle doses of Yixin Oral Liquid significantly slowed ligation at some time points. The elevation of left ventricular end-diastolic pressure (LVEDP) was elevated (P < 0.05), but the improvement of coronary flow (CF) was not significant. 2 Large and medium dose pretreatment with Yixin Oral Liquid reduced the decrease of SOD activity and the increase of MDA content in ischemic myocardium (P<0.05). There was no significant difference in the effect between pretreatment with Yixin Oral Liquid and diltiazem group and Compound Danshen Dripping Pill group. (P>0.05). 3 Yixin oral liquid large and medium doses and Compound Danshen Dripping Pills can relieve the attenuation of ATP content in ischemic myocardium (P<0.05). Conclusion Pretreatment with Yixin Oral Liquid can protect against ischemic injury of isolated rat myocardium and is dose-dependent. Its protective effect may involve antioxidative damage and effective preservation of energy.
