目的:检测肿瘤细胞条件培养液中VEGF的表达量。方法:在表达及分离纯化VEGF的基础上,成功地制备了VEGF抗体,以夹心法测定VEGF。结果:VEGF测定范围为002~20000ng/ml,灵敏度可达002ng/ml,批内及批间变异均小于10%,回收率平均为1024%。其中人胃癌细胞MGC803、BGC823、乳癌MCF7及肝癌BEL7402分泌上清中VEGF的含量分别为650、045、090及145ng/ml。结论:不同的肿瘤细胞系其VEGF的分泌量是不同的,应用此方法可以检测肿瘤病人血清中的VEGF。
Objective: To detect the expression of VEGF in conditioned medium of tumor cells. Methods: Based on the expression and purification of VEGF, VEGF antibody was successfully prepared and VEGF was detected by sandwich method. Results: The detection range of VEGF was 0200 ~ 20000 ng / ml. The sensitivity was up to 002 ng / ml. The variation of intra-assay and inter-assay were less than 10% and the average recovery was 1024%. Among them, the content of VEGF in human gastric cancer cells MGC803, BGC823, breast cancer MCF7 and liver cancer BEL7402 secretion supernatant were 650,045,090 and 1.45ng / ml, respectively. CONCLUSION: The secretion of VEGF in different tumor cell lines is different. This method can be used to detect the serum VEGF in tumor patients.
