目的：检测肿瘤细胞条件培养液中ＶＥＧＦ的表达量。方法：在表达及分离纯化ＶＥＧＦ的基础上，成功地制备了ＶＥＧＦ抗体，以夹心法测定ＶＥＧＦ。结果：ＶＥＧＦ测定范围为００２～２００００ｎｇ／ｍｌ，灵敏度可达００２ｎｇ／ｍｌ，批内及批间变异均小于１０％，回收率平均为１０２４％。其中人胃癌细胞ＭＧＣ８０３、ＢＧＣ８２３、乳癌ＭＣＦ７及肝癌ＢＥＬ７４０２分泌上清中ＶＥＧＦ的含量分别为６５０、０４５、０９０及１４５ｎｇ／ｍｌ。结论：不同的肿瘤细胞系其ＶＥＧＦ的分泌量是不同的，应用此方法可以检测肿瘤病人血清中的ＶＥＧＦ。 Objective: To detect the expression of VEGF in conditioned medium of tumor cells. Methods: Based on the expression and purification of VEGF, VEGF antibody was successfully prepared and VEGF was detected by sandwich method. Results: The detection range of VEGF was 0200 ~ 20000 ng / ml. The sensitivity was up to 002 ng / ml. The variation of intra-assay and inter-assay were less than 10% and the average recovery was 1024%. Among them, the content of VEGF in human gastric cancer cells MGC803, BGC823, breast cancer MCF7 and liver cancer BEL7402 secretion supernatant were 650,045,090 and 1.45ng / ml, respectively. CONCLUSION: The secretion of VEGF in different tumor cell lines is different. This method can be used to detect the serum VEGF in tumor patients.
