目的:体外研究姜黄素抑制活化的人脐静脉内皮细胞增殖及其与中性粒细胞黏附、迁移的影响,揭示前期动物实验姜黄素抑制角膜新生血管生长的机制。方法:四甲基偶氮噻唑蓝(MTT)法检测不同浓度姜黄素对AngⅡ活化的人脐静脉内皮细胞增殖的影响;观察分析其对活化的内皮细胞与中性细胞粘附率和迁移率的影响。结果:恒定的AngⅡ浓度活化的脐静脉内皮细胞的增殖及其与中性粒细胞的黏附率和迁移率被不同浓度姜黄素干预后其受到抑制并呈浓度依赖性,各不同浓度组与未加干预组比较差异有显著性,P=0.000。结论:姜黄素抑制AngⅡ活化的人脐静脉内皮细胞增殖及其与中性粒细胞的粘附与迁移,可能是其干预角膜新生血管生成的机制之一。
OBJECTIVE: To study the effect of curcumin on the proliferation of human umbilical vein endothelial cells and its effect on neutrophil adhesion and migration in vitro, and to reveal the mechanism by which curcumin inhibits corneal neovascularization in vitro. Methods: The effects of different concentrations of curcumin on the proliferation of human umbilical vein endothelial cells stimulated by Ang Ⅱ were detected by MTT assay. The effects of curcumin on the proliferation of endothelial cells and neutral cells were observed influences. RESULTS: Proliferation of human umbilical vein endothelial cells activated by constant AngⅡ concentration and its adhesion and migration rate with neutrophils were inhibited by concentration-dependent curcumin. Concentration-dependent, The difference between the intervention group was significant, P = 0.000. CONCLUSION: Curcumin inhibits the proliferation of human umbilical vein endothelial cells activated by Ang Ⅱ and its adhesion and migration with neutrophils, which may be one of the mechanisms of its intervention on corneal neovascularization.
