目的探讨辛伐他汀对糖基化终末产物(AGEs)诱导内皮细胞环氧化酶2(COX-2)表达的影响。方法体外以不同浓度的糖基化白蛋白(AGE-BSA)、辛伐他汀单独或联合作用于人脐静脉内皮细胞,检测细胞COX-2mRNA的表达及培养上清液中前列腺素E2(PGE2)的含量。结果 AGEBSA诱导内皮细胞COX-2mRNA的表达,作用呈剂量和时间依赖方式。辛伐他汀可降低AGE-BSA诱导的COX-2mRNA表达及PGE2产物浓度。结论 AGEs促进内皮细胞COX-2表达,辛伐他汀可减轻此作用。
Objective To investigate the effect of simvastatin on the expression of cyclooxygenase 2 (COX-2) induced by advanced glycation end products (AGEs). Methods Human umbilical vein endothelial cells were treated with AGE-BSA and simvastatin at different concentrations in vitro. The expression of COX-2 mRNA and the contents of prostaglandin E2 (PGE2) Content. Results AGEBSA induced COX-2 mRNA expression in endothelial cells in a dose-and time-dependent manner. Simvastatin can reduce AGE-BSA-induced COX-2 mRNA expression and PGE2 product concentration. Conclusion AGEs promote the expression of COX-2 in endothelial cells and simvastatin can reduce this effect.
