目的:建立测定连翘中芦丁和连翘酯苷含量的方法。方法:采用HPLC法。色谱柱为Thermo ODS C_(18)(250 mm×4.6 mm,5μm),流动相:甲醇-0.2%冰醋酸溶液(20∶80),流速1 ml·min~(-1),检测波长:329 nm,柱温30℃。结果:芦丁、连翘酯苷的进样量分别在0.023 2~0.464μg(r=0.999 9)和0.050 6~1.012μg(r=0.999 9)范围内呈良好线性关系;平均回收率分别为101.6%(RSD=1.38%,n=6)和100.8%(RSD=1.26%,n=6)。结论:本方法简便、准确,适用于连翘的分析测定和质量控制。
Objective: To establish a method for the determination of rutin and forsythiaside in Forsythia suspensa. Methods: HPLC method was used. The chromatographic column was Thermo ODS C18 (250 mm × 4.6 mm, 5 μm). The mobile phase was methanol-0.2% glacial acetic acid (20:80) at a flow rate of 1 ml · min -1. The detection wavelength was 329 nm, column temperature 30 ℃. Results: The injection volume of rutin and forsythiaside showed a good linearity in the range of 0.023 2-0.464 μg (r = 0.999 9) and 0.050 6-1.012 μg (r = 0.999 9) respectively. The average recoveries were 101.6% (RSD = 1.38%, n = 6) and 100.8% (RSD = 1.26%, n = 6). Conclusion: This method is simple and accurate, suitable for the determination of Forsythia and quality control.
