目的:确定鸟氨酸脱羧酶(ODC)反义寡核苷酸对HL60细胞的抗增殖作用;探索该核苷酸是否有可能作为一种治疗癌症的潜化制剂。方法:采用自行设计人工合成的ODC反义寡核苷酸,用MTT法观察了其对HL60细胞生长的影响,并以3H参入试验检测其RNA及蛋白生物合成。结果:ODC反义寡核苷酸可抑制HL60细胞的增殖及RNA、蛋白质的生物合成,并增强该细胞对抗肿瘤药物阿糖胞苷的敏感性。结论:应用反义技术控制ODC表达将是抑制恶性细胞生长及增强肿瘤对化疗敏感性的1种可能途径。
OBJECTIVE: To determine the anti-proliferative effect of ornithine decarboxylase (ODC) antisense oligonucleotides on HL60 cells and to explore whether this nucleotide might be used as a potential therapeutic agent for cancer. METHODS: Self-designed ODC antisense oligonucleotides were designed and their effects on the growth of HL60 cells were observed by MTT assay. RNA and protein biosynthesis were detected by 3H assay. RESULTS: ODC antisense oligonucleotides inhibited the proliferation of HL60 cells, RNA and protein biosynthesis, and enhanced the sensitivity of the cells to antitumor drug cytarabine. Conclusion: The use of antisense technology to control the expression of ODC is a possible way to inhibit the growth of malignant cells and enhance the sensitivity of tumors to chemotherapy.
