Gle1蛋白是一种mRNA核输出因子.最近有研究报道Gle1蛋白参与了酿酒酵母的蛋白质翻译终止过程.为了探讨Gle1蛋白是否在其它生物中也具有同样的功能,本研究利用酵母双杂交、免疫共沉淀以及免疫共定位等方法证实了人Gle1蛋白与参与蛋白质合成终止的两类肽链释放因子均能够相互作用,提示hGle1蛋白可能在人细胞中参与了蛋白质的翻译终止过程.进一步在HeLa细胞中利用双荧光素酶报告系统分析人Gle1蛋白对蛋白质翻译终止的影响,结果显示,hGle1的过表达能促进细胞内蛋白质的翻译终止.为进一步探讨hGle1蛋白在蛋白质合成中的作用机制奠定了基础.
Gle1 protein is an mRNA nuclear export factor.Recent studies have reported Gle1 protein involved in the termination of protein translation in S. cerevisiae.To explore whether Gle1 protein has the same function in other organisms, this study used yeast two-hybrid, Precipitation and co-immunophenotyping confirmed that human Gle1 protein interacts with two types of peptide chain release factors involved in the termination of protein synthesis, suggesting that hGle1 protein may be involved in the process of protein translation termination in human cells. In HeLa cells The dual luciferase reporter system was used to analyze the effect of human Gle1 protein on protein translation termination. The results showed that over-expression of hGle1 can promote the termination of protein translation in human cells, which laid a foundation of further study on the mechanism of hGle1 protein in protein synthesis.
