生物活性酶的固定化是几乎所有类型的生物传感器制备过程中必经的步骤。固定化酶的质量,除与生物活性物质本身有关外,固定化技术是一项重要的因素。本文以葡萄糖氧化酶(GOD)为对象,首次利用活蚕液状丝素蛋白的构象变化,即蛋白质的变性现象,将酶固定于蚕丝素蛋白中。用这种新的酶固定化方法所制成的葡萄糖氧化酶传感器,不仅具有制作方便,酶活性降低少,电极稳定性高,响应速度快,寿命长等特点,而且呈现优越的耐热能力。同时,对于同一批制备的酶——丝素蛋白柞品,所制电极的性能有着相当好的重现性,因而具有商品化应用前景。
Immobilization of bioactive enzymes is a necessary step in the preparation of almost all types of biosensors. The quality of immobilized enzyme, in addition to the biological activity itself, the immobilization technology is an important factor. In this paper, glucose oxidase (GOD) as the object, for the first time using the silkworm silk fibroin liquid conformational changes, that is, protein denaturation, the enzyme immobilized in silk fibroin protein. The glucose oxidase sensor made by the new enzyme immobilization method not only has the advantages of convenient manufacture, less decrease of enzyme activity, high stability of the electrode, fast response and long service life, etc., but also has the superior heat resistance capability. At the same time, for the same batch of prepared enzyme - silk fibroin oak products, the performance of the electrode has a very good reproducibility, which has the commercial application prospects.
