目的:弥补单纯珊瑚无骨诱导活性、骨修复能力弱等缺陷,给临床提供一种良好的骨移植替代材料。方法:将珊瑚与胶原和rhBMP-2复合,制备出珊瑚/胶原/rhBMP-2复合人工骨,将其植入大鼠背部肌肉陷窝内,以珊瑚/胶原和珊瑚/rhBMP-2复合人工骨植入作对照;取材后通过组织学方法和图像分析评价其骨诱导活性。结果:珊瑚/胶原/rhBMP-2植入后1周,在局部诱导出软骨细胞分化和软骨基质形成;4周形成含骨髓的板层骨;诱导成骨的量有明显的rhBMP-2剂量依赖性(P<0.01);而珊瑚/胶原和珊瑚/rhBMP-2植入区均无骨或软骨形成。结论:珊瑚/胶原/rhBMP-2复合人工骨具有良好的异位诱导成骨活性,是一种较理想的骨移植替代材料。
OBJECTIVE: To make up for the defects of pure coral, such as osteoinductive activity and weak ability of bone repair, to provide a good alternative material for bone transplantation. METHODS: Coral / collagen / rhBMP-2 composite artificial bone was prepared by compounding collagen with rhBMP-2 and implanted into the lacunae of the back muscles of rats. Coral / collagen and rhBMP-2 composite artificial bone Implantation as a control; After the material was drawn by histological methods and image analysis to evaluate the osteoinductive activity. RESULTS: One week after coral / collagen / rhBMP-2 implantation, chondrocyte differentiation and cartilage matrix formation were induced locally; bone marrow-containing lamellar bone was formed at 4 weeks; and the amount of osteoblasts induced by rhBMP-2 was significantly dose dependent (P <0.01). However, there was no bone or cartilage formation in the coral / collagen and coral / rhBMP-2 implants. CONCLUSION: Coral / collagen / rhBMP-2 composite artificial bone has good ectopic osteogenic activity and is an ideal substitute for bone graft.
