以表位为基础的基因免疫 (EBGI)是基因免疫技术发展的新方向 ,因其抗原性特定、无关干扰减少、效果更专一和精确而独具优势。影响EBGI效果的因素有表位的组建方式、表位的侧翼序列和辅助因子的融合 /共注射等。就此 ,若干增强EBGI技术的新策略已被提出和应用。就CTL表位而言 ,可通过在表位氨基端添加信号肽、氨基端融合蛋白酶体相关蛋白、表位与MHCI类分子偶联以及与 β2 m分子偶联等来增强其基因免疫效果。
Epitope-based genetic immunization (EBGI) is a new direction for the development of genetic immunization technology, because of its antigenic specificity, reduce irrelevant interference, the effect is more specific and precise and unique advantages. The factors influencing the efficacy of EBGI are the formation of epitopes, the flanking sequences of epitopes and the fusion / co-injection of cofactors. In this regard, several new strategies for enhancing EBGI technology have been proposed and applied. In the case of CTL epitopes, the effect of gene immunization can be enhanced by adding a signal peptide at the amino terminus of the epitope, an amino-terminal fusion proteasome-related protein, an epitope coupled with an MHC class I molecule, and coupling with a β2 m molecule.
