目的观察宫颈癌细胞系HeLa细胞在膜联蛋白A5表达降低后,细胞增殖和凋亡的情况。方法采用RNA干扰的方法,通过免疫印迹法筛选出对HeLa细胞中膜联蛋白A5的表达抑制效率最高的siRNA,脂质体法将siRNA转染入细胞48h后,用MTT法检测细胞增殖的改变情况,用细胞凋亡光学检测试剂盒检测细胞的凋亡。结果HeLa细胞中膜联蛋白A5的表达被显著抑制,其增殖速度与未转染siRNA的普通HeLa细胞相比,无显著性差异;细胞凋亡显著减少,与未转染siRNA的普通HeLa细胞相比,有显著性差异。结论膜联蛋白A5低表达对宫颈癌细胞系HeLa细胞的增殖未见显著影响,但可能影响了细胞的凋亡。
Objective To observe the proliferation and apoptosis of cervical cancer cell line HeLa after the expression of annexin A5 is decreased. Methods RNA interference was used to screen the siRNA with the highest inhibitory effect on Annexin A5 expression in HeLa cells. The siRNA was transfected into the cells for 48 h by lipofectamine and the cell proliferation was detected by MTT assay In the condition, apoptosis was detected by using the apoptosis detection kit. Results The expression of Annexin A5 in HeLa cells was significantly inhibited, and the proliferation rate of HeLa cells was not significantly different from that of the normal HeLa cells without transfected with siRNA. The apoptosis of HeLa cells was significantly decreased. Compared with the normal HeLa cells without siRNA Than, there are significant differences. Conclusion The low expression of Annexin A5 has no significant effect on the proliferation of cervical cancer cell line HeLa cells, but may affect the cell apoptosis.
