目的:探讨核糖核酸酶抑制因子(RNasin)对脑胶质瘤生长的抑制作用及可能机理。方法:从人胎盘组织中提取制备RNasin,体内外观察其对人SHG44和大鼠C6胶质瘤细胞生长的抑制作用。结果:(1)体外不同浓度的RNasin对培养的C6及SHG44胶质瘤细胞存活率无明显影响,与对照组比较无显著性差异(P>0.05);(2)加入RNasin后裸鼠体内C6胶质瘤平均重量为1.72 g±0.184 g,平均大小(直径)为12.84 mm±1.15 mm;SHG44胶质瘤平均重量为1.012 g±0.174 g,平均大小为11.20 mm±1.21mm,均与对照组有显著性差异(P<0.05)。结论:RNasin体外对培养的胶质瘤细胞生长无抑制作用,而体内对胶质瘤瘤体的生长有明显抑制作用。
Objective: To investigate the inhibitory effect and possible mechanism of ribonuclease inhibitor (RNasin) on glioma growth. METHODS: RNasin was extracted from human placenta tissue and its inhibitory effect on the growth of human SHG44 and rat C6 glioma cells was observed in vitro and in vivo. Results: (1) Different concentrations of RNasin in vitro had no significant effect on the survival rate of cultured C6 and SHG44 glioma cells, and there was no significant difference compared with the control group (P>0.05); (2) Nude mice after adding RNasin The average weight of C6 glioma in vivo was 1.72 g ± 0.184 g, the average size (diameter) was 12.84 mm ± 1.15 mm; the average weight of SHG44 glioma was 1.012 g ± 0.174 g The average size was 11.20 mm±1.21 mm, which was significantly different from the control group (P<0.05). Conclusion: RNasin has no inhibitory effect on the growth of cultured glioma cells in vitro, but it can significantly inhibit the growth of glioma tumors in vivo.
