小麦与玉米杂交产生的单倍体植株无染色体自然加倍发生 ,为获得加倍单倍体 ,进行了两种秋水仙素加倍处理。结果表明 ,在培养基中直接加入秋水仙素处理已培养 7d的幼胚 ,无秋水仙素对照的幼胚萌发率为 6 7.4 % ,存活的植株均未结实 ;而经过浓度为 5 0mg/L、10 0mg/L和 2 0 0mg/L秋水仙素处理的幼胚萌发率分别是 32 .1%、2 6 .4 %和 16 .3% ,加倍率分别是 85 .7%、10 0 %和 5 0 %。秋水仙素处理时间在 2 4~ 72h范围内 ,随着处理时间的延长 ,幼胚萌发率随之降低 ,死亡率随之增加 ,以 2 4h效果较好。用浓度为 5 0 0mg/L、75 0mg/L、10 0 0mg/L的秋水仙素溶液浸根加倍处理 ,平均加倍率分别是 89.6 %、76 .0 %和 73.3% ,而用浓度为 5 0 0mg/L秋水仙素处理壮苗获得了98.2 %的加倍率和 93.2 %的加倍处理效率
