目的检测肿瘤细胞中RNA结合蛋白Ro60的表达定位,研究Ro60在肿瘤增殖和辐射敏感性中的作用。方法构建带绿色荧光素酶标签的Ro60真核表达载体,转染肿瘤细胞,观察电离辐射引发的DNA损伤反应中Ro60的表达定位规律;CCK8和锥虫蓝染色实验检测Ro60蛋白对肿瘤细胞增殖和辐射敏感性的影响。结果多种肿瘤细胞中均有较高水平的Ro60蛋白表达。γ射线诱导的DNA损伤反应中Ro60发生显著的表达上调和亚细胞定位改变。过表达Ro60的肿瘤细胞增殖延缓,细胞死亡明显增多,辐射引发的细胞增殖抑制和死亡显著增强。结论辐射引发肿瘤细胞中Ro60表达和定位改变,调控肿瘤细胞增殖和凋亡,提高肿瘤细胞的辐射敏感性。
Objective To detect the expression and location of RNA binding protein Ro60 in tumor cells and study the role of Ro60 in tumor proliferation and radiosensitivity. Methods The Ro60 eukaryotic expression vector with green fluorescent luciferase was constructed and transfected into tumor cells. The expression and localization of Ro60 in DNA damage induced by ionizing radiation were observed. The expression of Ro60 was detected by CCK8 and Trypan blue staining. Radiation sensitivity effects. The results of a variety of tumor cells have a higher level of Ro60 protein expression. Ro60 induced a significant upregulation of expression and subcellular localization in γ-ray-induced DNA damage response. Proliferation of tumor cells over-expressing Ro60 was delayed, cell death was significantly increased, radiation-induced inhibition of cell proliferation and death increased significantly. Conclusion The radiation-induced Ro60 expression and localization in tumor cells can regulate the proliferation and apoptosis of tumor cells and enhance the radiosensitivity of tumor cells.
