目的 探讨白介素 1α(IL 1α)和白介素 1受体拮抗剂 (IL 1ra)对培养的Graves’眼病 (GO)患者眼眶成纤维细胞增殖的影响。方法 以下述方法分别处理培养的GO和正常人眼眶成纤维细胞 :不同浓度的IL 1α刺激成纤维细胞48h ;3 0U/ml的IL 1α刺激成纤维细胞 2 4~ 72h ;不同浓度的IL 1ra或 /和 3 0U/ml的IL 1α处理成纤维细胞 48h ;75ng/ml的IL 1ra和 3 0U/ml的IL 1α处理成纤维细胞 2 4~ 72h。用MTT法测定成纤维细胞增殖的刺激或抑制率。结果 IL 1α以剂量和时间依赖的方式刺激GO和正常人眼眶成纤维细胞增殖 ,而IL 1ra以剂量和时间依赖的方式抑制IL 1α诱导的GO和正常人眼眶成纤维细胞的增殖活性 ,刺激和抑制作用在GO和正常人眼眶成纤维细胞之间无明显差异。结论 IL 1ra是IL 1α诱导的人眼眶成纤维细胞增殖的强有力的抑制剂 ,可望用于GO的治疗
Objective To investigate the effects of interleukin-1α (IL-1α) and interleukin-1 receptor antagonist (IL-1ra) on proliferation of orbital fibroblasts in cultured Graves’ ophthalmopathy (GO) patients. Methods Cultured GO and normal human orbital fibroblasts were treated with different concentrations of IL-1α for 48h, fibroblasts stimulated with 30μL / ml IL-1αfor 24-72h, different concentrations of IL-1ra or Fibroblasts were treated with IL-1α at 30 U / ml for 48h, IL-1 at 75ng / ml and IL-1α at 30U / ml for 24-48 h. The stimulation or inhibition rate of fibroblast proliferation was measured by MTT method. Results IL 1α stimulated proliferation of GO and normal human orbital fibroblasts in a dose-and-time-dependent manner, whereas IL 1ra inhibited IL 1α-induced orbital fibroblast proliferation and proliferation in GO and normal human eyes in a dose- and time-dependent manner No significant difference in inhibition between GO and normal human orbital fibroblasts. Conclusions IL 1ra is a potent inhibitor of IL-1α-induced proliferation of human orbital fibroblasts and is expected to be used in the treatment of GO
