目的:探讨放射损伤对肠粘膜免疫的影响。方法:小鼠全身照射12Gy为模型,于照后8、24、48h取材光电镜观察,同时分离全小肠上皮内淋巴细胞(IEL),检测其细胞增殖活力、细胞毒力,并检测IEL培养上清中TNF-a、TGF-β浓度的变化。结果:①12Gy全身照射引起IEL数量减少,变性坏死的IEL能够造成相紧贴的肠上皮细胞损害。②12Gy全身照射后IEL增殖活力和细胞毒力下降。③12Gy全身照射后IEL分泌TNF-a、TGF-β增多。结论:12Gy全身照射引起肠粘膜免疫功能下降,导致肠道屏障功能削弱。
Objective: To investigate the effects of radiation injury on intestinal mucosal immunity. Methods: Whole body irradiation 12Gy was used as a model. After 8,24,48 hours of irradiation, the cells were observed under light microscope and the whole intestinal intraepithelial lymphocytes (IEL) were isolated. The viability and cytotoxicity of the cells were detected, and IEL culture Changes in serum TNF-a, TGF-β concentrations. Results: ①12Gy whole body irradiation caused a decrease in the number of IEL, degeneration and necrosis of IEL can cause close adhesion of intestinal epithelial damage. ② After 12Gy whole body irradiation IEL proliferative activity and cytotoxicity decreased. ③12Gy after IEL secretion of TNF-a, TGF-β increased. CONCLUSION: The systemic immunosuppression of 12 Gy causes a decrease in intestinal mucosal immunity, leading to impaired intestinal barrier function.
