目的:探讨三氧化二砷(arsenous oxide,As2O3)对胰腺癌PC2细胞的抑制作用及其作用机制。方法:用胰腺癌PC2细胞株进行培养传代,观察As2O3作用于细胞的不同时间,对细胞生长抑制情况;利用TUNEL染色检测胰腺癌细胞凋亡率;RT-PCR方法检测survivin基因的表达。结果:①随着药物对胰腺癌细胞作用时间的延长,细胞生存率明显降低,药物作用存在时间依赖性(P<0·05,P<0·01)。②TUNEL染色证实,As2O3对胰腺癌细胞的抑制作用是以促进细胞凋亡为主。③凋亡抑制因子survivin基因在胰腺癌细胞中表达;胰腺癌细胞经As2O3作用后,抑制sur-vivin基因的表达(P<0·05)。结论:As2O3可能通过抑制凋亡抑制因子survivin基因的表达,促进细胞凋亡,从而发挥抗肿瘤的作用。
Objective: To investigate the inhibitory effect of arsenous oxide (As2O3) on pancreatic cancer PC2 cells and its mechanism. Methods: Pancreatic cancer PC2 cells were used to culture and passage. The effects of As2O3 on cell growth were observed at different time points. The apoptosis rate of pancreatic cancer cells was detected by TUNEL staining and the expression of survivin gene was detected by RT-PCR. Results: ① With the prolongation of the effect of drugs on pancreatic cancer cells, the cell survival rate was significantly reduced, and the drug effect was time-dependent (P <0.05, P <0.01). ②TUNEL staining confirmed that the inhibitory effect of As2O3 on pancreatic cancer cells is mainly to promote cell apoptosis. The expression of survivin gene in pancreatic cancer cells was inhibited by As2O3, and the expression of survivin gene was inhibited (P <0.05). Conclusion: As2O3 may exert anti-tumor effect by inhibiting the expression of survivin gene and promoting cell apoptosis.
