目的研究白虎二地汤对实验性2型糖尿病(T2DM)胰岛素抵抗大鼠血糖、血脂的影响,并探讨其机制。方法采用高糖高脂饲料喂养联合低剂量链脲佐菌素腹腔注射建立T2DM胰岛素抵抗大鼠模型,将大鼠分为正常对照组、模型组、罗格列酮组、白虎二地汤高、低剂量组。治疗前后检测空腹血糖(FBG)、空腹胰岛素(FINS)、胰岛素抵抗指数(HOMA-IR)、血清总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)、游离脂肪酸(FFA)。显微镜观察五组大鼠肝脏、骨骼肌、脂肪的病理变化。结果白虎二地汤可明显降低2型糖尿病大鼠FBG、FINS、HOMA-IR及血清TC、TG、LDL-C、FFA水平,升高血清HDL-C浓度,减少肝脏组织脂肪浸润,且以高剂量白虎二地汤组效果最佳。结论白虎二地汤可以改善STZ诱导的T2DM胰岛素抵抗大鼠糖、脂代谢异常,减轻胰岛素抵抗。其可能的机制是通过降低脂毒性,增强胰岛组织对胰岛素的敏感性,发挥其改善糖脂代谢紊乱和降糖降脂作用。
Objective To study the effect of Baihu Erdi decoction on blood glucose and blood lipid in experimental type 2 diabetes mellitus (T2DM) rats with insulin resistance and to explore its mechanism. Methods High glucose and high fat diet combined with low dose of streptozotocin intraperitoneally injected T2DM insulin resistance rat model was established, the rats were divided into normal control group, model group, rosiglitazone group, Baihu Erdian Tang high, Low dose group. Fasting blood glucose, fasting insulin, HOMA-IR, TC, TG, HDL-C, Density lipoprotein cholesterol (LDL-C), free fatty acids (FFA). The pathological changes of liver, skeletal muscle and fat in the five groups were observed with microscope. Results Baihu Erdi Decoction could significantly decrease FBG, FINS, HOMA-IR, TC, TG, LDL-C and FFA in type 2 diabetic rats, increase serum HDL-C concentration and decrease fatty infiltration in liver tissues, White tiger two doses of the best dose group. Conclusion Baihu Erdi Decoction can improve STZ-induced glucose and lipid metabolism in T2DM rats with insulin resistance and reduce insulin resistance. Its possible mechanism is to reduce lipid toxicity and enhance insulin sensitivity of pancreatic islets to play its role in improving glucose and lipid metabolism disorders and hypoglycemic and lipid-lowering effect.
