中药提取液对弹性蛋白酶的抑制作用(体外实验)

蛋白酶-抗蛋白酶平衡失调在COPD发病中起重要作用。弹性蛋白酶(ELT)活性增高,可能是上述失衡的主要机制。寻找对ELT有抑制作用的药物,特别是从祖国中草药中发掘对防治COPD具有重要意义。山豆根、连翘、甘草等中药分别经水煎,乙醇提取,干燥,最后加水稀释成1ml含生药2.0g备用;常规制备琼脂糖板(含1%琼脂,0.08弹性蛋白,0.02%SDS及0.05%NaN3),打孔,每孔加样20μl,置37℃温育24小时,测量各孔溶解环直径,反应酶 The imbalance of protease-antiprotease plays an important role in the pathogenesis of COPD.
