目的:建立快速测定人体尿样中福多司坦药物浓度的高效液相色谱(HPLC)法。方法:采用HPLC反相柱,流动相为20mmol.L-1醋酸钠溶液(含0.6%四氢呋喃,pH7.2)-乙腈(81.5∶18.5),荧光检测器激发波长338nm,发射波长450nm。结果:福多司坦人体尿样在0.25~80mg.L-1范围内与峰面积呈良好的线性关系(r=0.999 1);RSD为2.6%,平均回收率为100.8%(n=5)。结论:本方法快速,准确,精密度良好,可用于健康人体尿样中福多司坦含量的测定。
Objective: To establish a high performance liquid chromatography (HPLC) method for the rapid determination of the concentration of bardustan in human urine samples. Methods: The HPLC reversed-phase column was used. The mobile phase consisted of 20mmol.L-1 sodium acetate solution (containing 0.6% tetrahydrofuran, pH7.2) -acetonitrile (81.5:18.5). The fluorescence detector was excited at 338nm and emission wavelength was 450nm. Results: There was a good linear relationship between the concentration of furosemide and peak area in the range of 0.25 ~ 80 mg.L-1 (r = 0.999 1), the RSD was 2.6% and the average recovery was 100.8% (n = 5) . Conclusion: The method is rapid, accurate and accurate. It can be used to determine the content of bortololide in healthy human urine samples.
