目的探讨WASP家族Verprolin同源蛋白1(WAVE1)和高迁移率族蛋白-1(HMGB1)在兔尿源性脓毒血症肺组织中表达上调中的关系。方法 30只健康雄性家兔随机分为假手术组(Sham组)和尿源性脓毒血症组(US组)。采用肾盂内高压法制备家兔尿源性脓毒血症模型。免疫组织化学染色SABC法分别于术后12 h、24 h、36 h检测家兔肺组织WAVE1蛋白表达水平及肺组织中HMGB1含量及HMGB1 mRNA表达。结果与Sham组比较,US组肺组织HMGBl含量、HMGBl mRNA表达活性升高(P<0.05);免疫组织化学结果显示,US组肺组织中WAVE1 80.00%(12/15))呈强阳性表达,在Sham组肺组织中,WAVE1 86.67%(13/15))呈低表达或不表达,差异具有统计学意义(P<0.05)。结论尿源性脓毒血症急性肺损伤中WAVE1与HMGB1可能具有相关性。
Objective To investigate the relationship between WASP family members Verprolin homology protein 1 (WAVE1) and high mobility group box protein-1 (HMGB1) in lung tissue of rabbits with urinary sepsis. Methods Thirty healthy male rabbits were randomly divided into sham operation group (Sham group) and urinary sepsis group (US group). Rabbits with urinary sepsis model were prepared by intra-renal pelvic hypertension. Immunohistochemical SABC method was used to detect the protein expression of WAVE1 and the content of HMGB1 and the expression of HMGB1 mRNA in the lung tissue of rabbits at 12 h, 24 h and 36 h respectively. Results Compared with Sham group, the levels of HMGB1 and HMGB1 mRNA in lung tissue of US group were significantly increased (P <0.05). Immunohistochemistry showed that WAVE1 was strongly expressed in the lung tissue of US group (80.00%, 12/15) In the lung tissue of Sham group, WAVE1 86.67% (13/15)) showed low or no expression, the difference was statistically significant (P <0.05). Conclusions WAVE1 and HMGB1 may be correlated with acute urinary sepsis-induced acute lung injury.
