背景:神经生长因子治疗周围神经的损伤已经得到充分的研究并被认可,但其能否通过血脑屏障发挥作用一直不能得到肯定结论,试图以脂质体包裹神经生长因子,并考察其与单纯神经生长因子的对比,证明其能通过血脑屏障,进入中枢神经系统。目的:通过单光子发射计算机断层显像技术(singlephotonemissioncom-puterizedtomography,SPECT)对比分析不同剂型的神经生长因子经静脉注射后在体内的靶向分布情况。设计:以试验动物为研究对象的随机对照研究。单位:一所医科大学附属医院。材料:实验于2003-06/2004-05于南京森科医药公司,南京医科大学第一附属医院核医学科完成,选用健康新西兰兔3个月龄(19只),普通级,体质量(2.0±0.2)kg,南京安立默科技有限公司提供,随机分组。犤99Tcm犦-神经生长因子(南京森科医药公司帮助标记,标记率:98.9%,放化纯度为:99.7%);脂质体(沈阳药科大学提供);200g/L乌拉坦注射液(南京医科大学试剂科提供)。方法:将犤99Tcm犦-神经生长因子用脂质体包裹后,按照以下条件处理:将包裹含有1.48×108Bq犤99Tcm犦-神经生长因子的脂质体A注射至新西兰兔体内,利用SPECT分析其在脑部分布的百分数。其他两组以相同放射剂量的犤99Tcm犦-神经生长因子,犤99Tcm犦-神经生长因子-普通脂质体B处理。主要观察指标:?
BACKGROUND: Nerve growth factor (NGF) treatment of peripheral nerve injury has been well studied and accepted. However, whether it can function through the blood-brain barrier has not been able to reach a conclusion. OBJECTIVE: To compare and analyze the targeted distribution of nerve growth factor (NGF) in different dosage forms after intravenous injection by single photon emission computed-tomography (SPECT). Design: Randomized controlled study of experimental animals. Unit: a Medical University Hospital. MATERIALS: The experiment was performed at the Department of Nuclear Medicine, Nanjing Senke Pharmaceutical Co., Ltd. and the First Affiliated Hospital of Nanjing Medical University from June 2003 to May 2004. The healthy New Zealand rabbits were selected for three months (19 months), normal grade and 2.0 ± 0.2) kg, provided by Nanjing Anrmer Technology Co., Ltd., were randomly divided into groups.犤 99Tcm 犦 - nerve growth factor (Nanjing Senke Pharmaceutical Co., Ltd. to help mark, labeling rate: 98.9%, radiochemical purity: 99.7%); liposome (Shenyang Pharmaceutical University); 200g / L urethane injection Nanjing Medical University, provided by the reagent).
