目的:探讨余甘子总黄酮(PEF)对乳腺癌细胞MCF-7增殖及凋亡的影响。方法:采用SRB法检测PEF对MCF-7细胞增殖的影响,集落形成法检测PEF对MCF-7细胞集落形成能力的影响;采用Hoechst33258染色法观察PEF对肿瘤细胞凋亡的影响。结果:不同浓度PEF分别作用细胞24h、48h和72h后,药物对细胞增殖有不同程度的抑制作用,呈时间和剂量依赖性。6μg/m L和12μg/m LPEF组的集落抑制率分别为52.84%和43.1%。给予不同浓度PEF诱导后,细胞出现细胞核或细胞质中浓染的蓝色颗粒及细胞形态改变,凋亡率有显著差异。结论:PEF可显著抑制MCF-7细胞增殖并诱导细胞凋亡。
Objective: To investigate the effect of total flavonoids of Phyllanthus emblica (PEF) on proliferation and apoptosis of breast cancer cell line MCF-7. Methods: The effect of PEF on the proliferation of MCF-7 cells was detected by SRB method. The colony formation assay was used to detect the effect of PEF on the colony formation ability of MCF-7 cells. Hoechst33258 staining was used to observe the effect of PEF on the apoptosis of MCF-7 cells. Results: The cells treated with different concentrations of PEF for 24h, 48h and 72h, respectively, inhibited the proliferation of cells in a dose-dependent and time-dependent manner. The colony inhibition rates of 6μg / ml and 12μg / ml LPEF group were 52.84% and 43.1% respectively. After being given different concentrations of PEF, the cells appeared the blue granules in the nucleus or the cytoplasm, and the morphological changes of the cells were observed. The apoptotic rates were significantly different. Conclusion: PEF can significantly inhibit MCF-7 cell proliferation and induce apoptosis.
