目的观察艾烟冷凝物(PM10采取人体可吸入的艾烟部分)对肺泡Ⅱ型上皮细胞(A549)活性及凋亡的影响。方法体外培养A549细胞,加入不同浓度的艾烟冷凝物,应用四甲基偶氮唑蓝(MTT)法、荧光显微镜来观察其对A549的细胞活性以及细胞凋亡的影响。结果 MTT法结果显示:与对照组相比,A549的细胞存活率随浓度和时间发生改变,具有明显的时间浓度依赖性;0.12g/L浓度的艾烟冷凝物作用于细胞12 h后可显著提高细胞存活率(P=0.005<0.01);荧光显微镜下观察发现艾烟冷凝物可以引起细胞发生凋亡,且具有浓度依赖性。结论 A549细胞随着艾烟冷凝物浓度的增加、刺激时间的增加而引起细胞活力逐渐下降,表明一定浓度的艾烟冷凝物和一定的刺激时间对细胞具有毒性作用;一定浓度的艾烟冷凝物在较短刺激时间内具有细胞增殖作用,故认为艾烟对细胞的增殖作用可能是艾烟发挥有效作用的重要因素之一;能够引起细胞的凋亡可能是毒性作用的重要因素之一。
Objective To observe the effect of PM10 on the activity and apoptosis of alveolar type Ⅱ epithelial cells (A549) induced by inhalation of Ai smoke (PM10). Methods A549 cells were cultured in vitro, and different concentrations of Aconite condensate were added. The effects of different concentrations on the cell viability and apoptosis of A549 cells were observed by MTT assay and fluorescence microscopy. Results The results of MTT assay showed that the cell viability of A549 changed with time and concentration in a time-dependent manner. Compared with the control group, the cell viability of A549 cells in a time-dependent manner was significant. After exposure to 0.12 g / L of Acylate condensate for 12 h, (P = 0.005 <0.01). It was observed by fluorescence microscope that Ash condensate could cause cell apoptosis in a concentration-dependent manner. CONCLUSION: A549 cells, with the increase of the concentration of AI smoke and the increase of stimulation time, caused a gradual decrease in cell viability, indicating that certain concentrations of AI smoke condensate and certain stimulation time have a toxic effect on cells. A certain concentration of AI smoke condensate It has the function of cell proliferation in a short stimulation time, so it is considered that the proliferation of AY on the cells may be one of the important factors that Aiyan play an effective role in. Apoptosis may be one of the important factors of toxicity.
