目的建立二色补血草中杨梅苷、圣草酚、木犀草素、槲皮素4种化学成分含量测定的HPLC方法。方法采用Accurasil C18(4.6 mm×250 mm,5μm)色谱柱;0.2%磷酸溶液和乙腈进行梯度洗脱;柱温25℃;流速1 m L·min~(-1);检测波长254 nm。结果在HPLC法中,杨梅苷、圣草酚、木犀草素、槲皮素的线性范围分别是:0.2232~0.6696μg(r=0.9995)、0.2196~0.6588μg(r=0.9997)、0.0684~0.2052μg(r=0.9996)、0.0624~0.1872μg(r=0.9998);平均回收率分别为98.5%、97.8%、99.5%、99.7%,RSD值分别为1.3%、1.6%、0.8%、1.6%。结论所建立的方法专属性强,重复性好,可用于二色补血草的质量控制。
OBJECTIVE To establish HPLC method for the determination of 4 chemical constituents of Myricetin, Eriochrome, Luteolin and Quercetin in Dictyostelium. Methods Column with Accurasil C18 (4.6 mm × 250 mm, 5 μm), gradient elution with 0.2% phosphoric acid and acetonitrile was used. The column temperature was 25 ℃ and the flow rate was 1 m L · min -1. The detection wavelength was 254 nm. Results The linear ranges of myricetin, eriodictyol, luteolin and quercetin were 0.2232 ~ 0.6696μg (r = 0.9995), 0.2196 ~ 0.6588μg (r = 0.9997), 0.0684 ~ 0.2052μg (r = 0.9996) and 0.0624 ~ 0.1872μg (r = 0.9998). The average recoveries were 98.5%, 97.8%, 99.5% and 99.7%, respectively. The RSDs were 1.3%, 1.6%, 0.8% and 1.6% respectively. Conclusion The established method is highly specific and reproducible and can be used for the quality control of Limonium bicolor.
