目的探讨TNF-α-308基因单核苷酸多态性与乳腺癌易感性的关系。方法采用PCR限制片段长度多态性方法对原发性乳腺癌患者165例和对照组170例健康志愿者进行TNF-α编码基因的308基因位点多态性分析。结果两组TNF-α-308基因多态型和等位基因型分布频率差异无统计学意义(P>0.05)。以有无脉管侵犯、淋巴结转移状况等进行分层分析显示,携带G等位基因的乳腺癌患者脉管侵犯、淋巴结转移风险增高[(OR=2.63,95%CI(1.29~5.37),P=0.01;OR=2.34,95%CI(1.05~5.20),P=0.04]。结论携带G等位基因的女性乳腺癌患者发生脉管侵犯、淋巴结转移的风险较高。
Objective To investigate the relationship between single nucleotide polymorphism of TNF-α-308 gene and breast cancer susceptibility. Methods A total of 165 patients with primary breast cancer and 170 healthy volunteers were enrolled in this study. The polymorphism of 308 loci in TNF-α-encoding gene was analyzed by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). Results There was no significant difference in the frequency of TNF-α-308 gene polymorphism and allele distribution between the two groups (P> 0.05). Hierarchical analysis of vascular invasion and lymph node metastasis showed that the risk of vascular invasion and lymph node metastasis was increased in patients with G allele [(OR = 2.63, 95% CI 1.29-5.37), P = 0.01; OR = 2.34, 95% CI 1.05 ~ 5.20, P = 0.04] .Conclusion Female breast cancer patients with G allele have a higher risk of vascular invasion and lymph node metastasis.
