目的探讨赤芍总苷(TPG)对人黑色素瘤A375细胞血管内皮生长因子(VEGF)和促血管生成素-2(Ang-2)表达的影响。方法体外培养人黑色素瘤A375细胞,分别加入12.5、25、50、100、200mg/LTPG,对照组加入等量生理盐水,孵育24h、48h后,MTT方法测定生长抑制率;RT-PCR、Western blot分别测定TPG处理48h后VEGF和Ang-2 mRNA和蛋白的表达。结果 MTT结果显示TPG对黑色素瘤细胞增殖有显著抑制作用;RT-PCR、Western blot结果显示,TPG可显著抑制人黑色素瘤细胞VEGF、Ang-2 mRNA水平及蛋白水平的表达。结论 TPG具有显著的抗黑色素瘤瘤血管生成的作用,其机制可能与下调VEGF、Ang-2的表达有关。
Objective To investigate the effect of total paeoniflorin (TPG) on the expression of vascular endothelial growth factor (VEGF) and angiopoietin-2 (Ang-2) in human melanoma A375 cells. Methods Human melanoma A375 cells were cultured in vitro and treated with 12.5, 25, 50, 100 and 200 mg / L TPG respectively. The control group was treated with the same amount of normal saline. The growth inhibition rate was determined by MTT assay after 24h and 48h incubation. The expressions of VEGF and Ang-2 mRNA and protein were detected after 48 h treatment with TPG. Results MTT results showed that TPG significantly inhibited the proliferation of melanoma cells. The results of RT-PCR and Western blot showed that TPG significantly inhibited the expression of VEGF and Ang-2 mRNA and protein in human melanoma cells. Conclusion TPG has a significant anti-angiogenic effect on melanoma, which may be related to the down-regulation of the expression of VEGF and Ang-2.
