目的:研究护骨胶囊含药血清对大鼠成骨细胞增殖、分化及成骨能力的影响,并初步探讨其调节成骨细胞功能的可能机制。方法:酶消化法体外培养并鉴定大鼠成骨细胞,血清药理学方法制备护骨胶囊含药血清和空白血清对成骨细胞进行干预,CCK-8试剂盒和碱性磷酸酶试剂盒分别检测成骨细胞的增殖和分化情况,半定量RT-PCR法和Western blot法检测成骨功能指标基因和上皮细胞钠离子通道(ENa C)基因的m RNA和蛋白表达。结果:与空白血清组比较,5%、10%护骨胶囊含药血清可显著促进成骨细胞增殖(P<0.05),10%、20%浓度的护骨胶囊含药血清可显著促进成骨细胞分化(P<0.05),10%护骨胶囊含药血清可显著上调成骨功能指标基因和ENa C基因m RNA和蛋白的表达(P<0.01)。结论:护骨胶囊含药血清能明显提高成骨细胞的增殖、分化和骨形成能力,且同时上调了成骨细胞上ENa C基因的m RNA和蛋白表达,提示护骨胶囊可能通过ENa C介导的调节途径改善成骨细胞功能,为明确其药效机制提供了新思路。
OBJECTIVE: To study the effects of serum containing HuGu Capsule on the proliferation, differentiation and osteogenic ability of osteoblasts in rats and to explore its possible mechanism of regulating the function of osteoblasts. Methods: Rat osteoblasts were cultured and identified by enzymatic digestion. Osteoblasts were prepared by serum pharmacology method. Serum and blank serum of Prescription for Caregiver Capsule were used to interfere with osteoblasts. CCK-8 kit and alkaline phosphatase kit were tested respectively The proliferation and differentiation of osteoblasts were detected by semi-quantitative RT-PCR and Western blot. The mRNA and protein expressions of osteoblasts and sodium channel (ENaC) in epithelial cells were detected. Results: Compared with the blank serum group, 5% and 10% serums of the Prescription for the Prescription of the Prescription for the Prescription of Gugu Capsules significantly promoted the proliferation of osteoblasts (P <0.05). The serum containing 10% and 20% Cell differentiation (P <0.05). The drug serum containing 10% Hugu capsule could significantly upregulate the expression of osteogenic marker gene and ENa C mRNA and protein (P <0.01). CONCLUSION: The Herbal Medicine Capsule can significantly enhance the proliferation, differentiation and osteogenesis of osteoblasts and up-regulate the expression of m RNA and protein of ENa C gene on osteoblasts, suggesting that the Prescription Capsule may be administered by ENa C The regulatory pathway leads to the improvement of osteoblast function, which provides a new idea for clarifying its mechanism of action.
