目的观察硫化氢(H2S)/胱硫醚-γ-裂解酶(CSE)对肝细胞再生的影响。方法以43例慢性乙型肝炎患者为肝炎组,21例肝损害治疗后正常者的非肝炎患者作为对照组。采用肝穿刺获得两组肝组织,用免疫组化SP法检测肝组织中CSE表达。用胶原酶消化法分离两组肝细胞,分别给予促肝细胞生长素(HGF)50μg/L、CSE抑制剂炔丙基甘氨酸(PPG)2 mmol/L+HGF 50μg/L、H2S供体NaHS 1 mmol/L+HGF 50μg/L刺激24 h,用MTT法观察两组细胞增殖变化。结果肝炎组肝组织CSE平均光密度值为0.5019±0.103,对照组为1.691±0.513,肝炎组CSE平均光密度值较对照组低(P<0.05)。HGF刺激后,肝炎组肝细胞吸光度值高于对照组(P<0.05);加CSE抑制剂PPG后,吸光度值增加更明显(P<0.05);加NaHS后,两组细胞吸光度均减弱(P<0.05)。结论 H2S/CSE具有抑制肝细胞增殖作用,减少内源性H2S将有利于肝再生。
Objective To observe the effect of hydrogen sulfide (H2S) / cystathionine-γ-lyase (CSE) on hepatocyte regeneration. Methods Forty-three patients with chronic hepatitis B were hepatitis and 21 healthy controls were enrolled as control group. Two groups of liver tissues were obtained by liver puncture. The expression of CSE in liver tissues was detected by immunohistochemical SP method. Two groups of hepatocytes were isolated by collagenase digestion, and 50 μg / L HGF, 50 μg / L CSH inhibitor propargylglycine (PPG) and 50 μg / L HGF donor, respectively, mmol / L + HGF 50μg / L for 24 h, the cell proliferation was observed by MTT assay. Results The average CSE value of hepatic tissue in hepatitis group was 0.5019 ± 0.103, while that in control group was 1.691 ± 0.513. The mean optical density of CSE in hepatitis group was lower than that of control group (P <0.05). After HGF stimulation, the absorbance of hepatocytes in Hepatitis group was higher than that in control group (P <0.05). After adding CSE inhibitor PPG, the absorbance value increased more obviously (P <0.05) <0.05). Conclusions H2S / CSE can inhibit the proliferation of hepatocytes and reduce endogenous H2S will be beneficial to liver regeneration.
