本文把PHA和电离辐射两种因子,在单独和合并应用的条件下,观察了它们对大鼠胸腺和淋巴结细胞LDH同功酶表达所带来的影响。发现PHA在适宜剂量下,使T细胞LDH_5比例增高。但高剂量PHA对淋巴细胞的LDH_5或总的M型酶的比例均无明显影响。在经250rad~(50)Coγ射线照射后,淋巴细胞表现为LDH_1下降和LDH_5增高,但以LDH_1的下降最为突出,差别非常显著。从亚基分析来看,电离辐射主要使H型酶的比例下降。高剂量PHA虽未见改变正常淋巴细胞的LDH同功酶的组成与谱带分布,但具有明显地防止辐射所引起的LDH_1或H型酶显著下降和LDH_5或M型酶增高的作用。它提示辐射对淋巴细胞的生物效应,受到细胞所处功能状态的影响。这对进一步研究外界刺激的作用与免疫潜能细胞功能状态之间的关系,也有一定参考价值。
In this paper, the effects of PHA and ionizing radiation on the expression of LDH isoenzymes in thymus and lymph node cells of rats were observed under the conditions of single and combined application. PHA was found to increase the proportion of LDH_5 T cells at a suitable dose. However, high-dose PHA had no significant effect on the proportion of LDH_5 or total M-type enzymes in lymphocytes. After exposure to 250 rad~(50)Co γ-rays, lymphocytes showed a decrease in LDH_1 and an increase in LDH_5, but the decrease in LDH_1 was most prominent, and the difference was significant. From the analysis of subunits, ionizing radiation mainly reduces the proportion of H-type enzymes. Although high-dose PHA did not change the composition and band distribution of LDH isoenzymes of normal lymphocytes, it had a significant effect of preventing radiation-induced LDH_1 or H-type enzymes from significantly decreasing and LDH_5 or M-type enzymes from increasing. It suggests that the biological effects of radiation on lymphocytes are affected by the functional status of the cells. This also has certain reference value for further studying the relationship between the external stimuli and the functional status of the immune potential cells.
