目的研究补肾中药仙鹿壮骨方对骨质疏松症大鼠骨组织I型胶原基因mRNA表达的影响。方法用地塞米松制造骨质疏松症大鼠模型,给予不同剂量的仙鹿壮骨方药,并与西药补钙剂对照观察。测定大鼠骨密度,以荧光定量PCR检测I型胶原基因表达水平。结果模型组骨密度明显下降,治疗各组骨密度均有提高。模型组I型胶原基因表达显著下降,治疗各组I型胶原基因表达明显上升,上升幅度以中药大剂量组最高,西药组次之,中药小剂量较差。结论补肾中药仙鹿壮骨方能促进地塞米松型骨质疏松症大鼠I型胶原的合成,其防治骨质疏松症可能是通过促进骨组织中I型胶原基因的表达,提高骨密度而实现的。
Objective To investigate the effect of Bushen Zhuanggu Zong Gu Fang on the expression of type I collagen mRNA in bone tissue of rats with osteoporosis. Methods Dexamethasone was used to make osteoporosis rats model, and different doses of Xianlu Zhuanggu Prescription were given and compared with Western medicine calcium supplements. Bone mineral density was measured in rats and the level of type I collagen gene expression was detected by fluorescent quantitative PCR. Results BMD decreased significantly in the model group, and bone mineral density was improved in all groups. The expression of type I collagen gene in the model group was significantly decreased, and the expression of type I collagen gene in each group was significantly increased. The increase was the highest in the high-dose Chinese medicine group, followed by the western medicine group, followed by the small dose of the Chinese medicine. CONCLUSION: Bushen Zhuanggu Zhuanggu can promote the synthesis of type I collagen in rats with dexamethasone-induced osteoporosis. The prevention and treatment of osteoporosis may be through the promotion of the expression of type I collagen gene in bone tissue and increase the bone density. Realized.
