本文研究旨在探索BB大鼠新生期刺激β细胞能否诱导耐受力,从而预防糖尿病的发生。 材料和方法 将BB大鼠配成对,新生大鼠产后立即作如下处理:每天2次(早晚)共6天,即腹腔注射100μl无菌用水,其中已先加入:0.01mg胰高血糖素加5mg葡萄糖;7.5mgL-精氨酸加2.5mg葡萄糖或20mg葡萄糖;对照组不作处理。3周后断奶,允许自由进食和饮水。每天观察动物,每周称体重并检查糖尿。如果出现至少++糖尿伴体重减轻10%以上(与前1周相比),则诊断为糖尿病。诊断一旦成立,处死动物测血糖。于第200天实验研究结束。
This study aims to explore BB neonatal stimulation of β cells can induce tolerance, thus preventing the occurrence of diabetes. MATERIALS AND METHODS BB rats were paired. Neonatal rats were treated immediately after childbirth as follows: 2 times a day (morning and evening) for 6 days, ie 100 μl of sterile water for peritoneal injection, of which 0.01 mg of glucagon plus 5 mg glucose; 7.5 mg L-arginine plus 2.5 mg glucose or 20 mg glucose; control group without treatment. Weaning after 3 weeks, allowing free access to food and water. Animals are observed daily, weighed weekly and checked for diabetes. Diabetes mellitus was diagnosed if at least ++ had a more than 10% weight loss with diabetics (compared to the previous week). Once diagnosed, animals were sacrificed to measure blood sugar. The experimental study was completed on the 200th day.
