目的:建立气道炎症模型豚鼠血浆中花生四烯酸(AA)的测定方法,考察野鸢尾黄素降低气道炎性反应的作用。方法:以烟熏配合合胞病毒滴鼻制造气道炎症豚鼠模型,以AA-d8为内标,采用Triton-14对血浆样品进行浊点萃取,并采用LC-MS/MS法MRM监测模式测定豚鼠血浆中AA的浓度。结果:血浆中AA在0.1~100μg·m L-1范围内线性关系良好,日内精密度为2.53%~7.82%,日间精密度为10.51%~13.41%,提取回收率81.20%~90.77%。结论:该方法环保、准确、稳定,可用于豚鼠血浆中AA的测定。
OBJECTIVE: To establish a method for the determination of arachidonic acid (AA) in guinea pigs with airway inflammation and to study the effect of iridoid on the airway inflammatory response. Methods: The guinea pig model of airway inflammation was made by intranasal administration of smoked complex syncytial virus. The plasma samples were extracted by Triton-14 with AA-d8 as internal standard and analyzed by LC-MS / MS MRM AA plasma concentrations in guinea pigs. Results: The linearity of AA in plasma ranged from 0.1 to 100μg · m L-1. The intra-day precision was 2.53% -7.82%, the intra-day precision was 10.51% -13.41%, and the extraction recovery was 81.20% -90.77%. Conclusion: The method is environmentally friendly, accurate and stable and can be used for the determination of AA in plasma of guinea pigs.
