目的:探讨缺氧缺血脑损伤(HIBD)对新生大鼠脑皮质中IL-6水平和Fas-L蛋白表达的影响。方法:建立HIBD大鼠模型后分为实验组、假手术组,于缺氧缺血或手术后1d、3d及7d,用放射免疫检测法测定脑皮质匀浆中IL-6的密度;用免疫组化染色法检测Fas-L的表达。结果:脑皮质组织匀浆中IL-6的浓度在缺氧缺血后1d、3d及7d,实验组大鼠依次为(56.4±7.2)ng/L、(250.4±11.3)ng/L及(345.8±12.4)ng/L。在相应的时间点,假手术组大鼠依次为(50.2±6.3)ng/L、(50.7±6.5)ng/L及(51.5±6.7)ng/L。缺氧缺血后3d及7d,IL-6的水平明显高于假手术组(P<0.05)。左脑皮质中表达Fas-L蛋白细胞的阳性率于缺氧缺血后1d、3d及7d,实验组大鼠依次为(12.21±6.1)%、(87.7±5.9)%及(8.33±2.9)%。在相应的时间点,假手术组大鼠依次为(8.41±3.5)%、(8.21±3.1)%及(8.02±2.8)%。缺氧缺血后1d及3d,实验组大鼠Fas-L的表达明显高于假手术组(P<0.05)。结论:缺血缺氧后一定时间内,可引起新生大鼠脑皮质中IL-6水平及Fas-L的表达明显增加,前者的持续时间较长,后者短暂。
Objective: To investigate the effect of hypoxic-ischemic brain damage (HIBD) on IL-6 and Fas-L protein expression in the cortex of neonatal rats. Methods: HIBD rats were divided into experimental group and sham operation group, and the density of IL-6 in cerebral cortex homogenate was detected by radioimmunoassay at 1d, 3d and 7d after hypoxia-ischemia or surgery; The expression of Fas-L was detected by histochemical staining. Results: The concentrations of IL-6 in cerebral cortex homogenate were (56.4 ± 7.2) ng / L, (250.4 ± 11.3) ng / L and 345.8 ± 12.4) ng / L. At the corresponding time points, the rats in the sham group were (50.2 ± 6.3) ng / L, (50.7 ± 6.5) ng / L and (51.5 ± 6.7) ng / L, respectively. The level of IL-6 was significantly higher than that of the sham operation group (P <0.05) 3d and 7d after hypoxia-ischemia. The positive rates of Fas-L protein expression in the left cerebral cortex were (12.21 ± 6.1)%, (87.7 ± 5.9)% and (8.33 ± 2.9)% in the order of 1d, 3d and 7d after hypoxic- %. At the corresponding time points, the rats in the sham operation group were (8.41 ± 3.5)%, (8.21 ± 3.1)% and (8.02 ± 2.8)%, respectively. At 1 and 3 days after hypoxia and ischemia, the expression of Fas-L in experimental group was significantly higher than that in sham-operated group (P <0.05). Conclusion: Within a certain period of time after ischemia and hypoxia, IL-6 and Fas-L expression in the cerebral cortex of neonatal rats were significantly increased. The former lasted longer and the latter short.
