目的探讨5-氟尿嘧啶(5-Fu)抗结肠癌作用中,细胞凋亡与P-gp(P-glycoprotein)蛋白表达的关系。方法应用琼脂培养MTT法,测定人结肠癌对抗代谢药的药物敏感性,同时观察5-Fu对结肠癌病人手术前化疗疗效;用S-P免疫组化和图像定量分析技术,测定34例人结肠癌组织中P-gp蛋白表达量;采用末端标记法(TUNEL),检测结肠癌细胞的凋亡指数。结果(1)P-gp蛋白表达量与5-Fu抗药指数呈正相关(r=0.675,P<0.05)。(2)凋亡率与5-Fu抗药指数呈负相关(r=-0.53,P<0.05)。(3)以5-Fu抗药指数1.0为界,>1为耐药组,<1为敏感组,两组间P-gp蛋白表达量、凋亡指数差异均有显著性(P<0.05)。结论5-Fu通过诱导细胞凋亡产生抗结肠癌作用,而P-gp蛋白高表达阻碍5-Fu诱导的凋亡。
Objective To investigate the relationship between apoptosis and the expression of P-glycoprotein in anti-colon cancer cells treated with 5-fluorouracil (5-Fu). Methods Agar cultured MTT method was used to determine the drug sensitivity of human colon cancer to antimetabolites. At the same time, the effect of 5-Fu on the preoperative chemotherapy of colon cancer patients was observed. SP immunohistochemistry and image quantitative analysis were used to detect 34 cases of human colon cancer The expression of P-gp protein in the tissue; The apoptosis index of colon cancer cells was detected by TUNEL. Results (1) The expression of P-gp protein was positively correlated with the 5-Fu drug resistance index (r = 0.675, P <0.05). (2) There was a negative correlation between apoptosis rate and 5-Fu resistance index (r = -0.53, P <0.05). (3) The 5-Fu resistance index of 1.0 for the sector,> 1 for the resistance group, <1 for the sensitive group, P-gp protein expression between the two groups were significantly different (P <0.05) . Conclusions 5-Fu has an anti-colon cancer effect by inducing apoptosis, while high expression of P-gp protein inhibits 5-Fu-induced apoptosis.
